Yastremska broke serve five times bouncing the top-seeded Spaniard out of Hua Hin, 7-6 (5), 6-1, to power into the Thailand Open semifinals.
The aggressive Ukrainian has been a nightmare opponent for Muguruza, sweeping all four sets they've played including a 6-2, 6-3, dismissal en route to the Luxembourg semifinals last fall.
In today's rematch, Yastremska pounded nearly six times as many winners (29 to 6) as the two-time Grand Slam champion.
It is Yastremska's second win over a former Grand Slam champion this season following her straight-sets win over former US Open champion Samantha Stosur in the opening round of the Australian Open.
The 47th-ranked Yastremska will play Magda Linette for a spot in Sunday's final.
Linette defeated Yafan Wang, 6-2, 6-3.
Australian Ajla Tomljanovic pounded out a 6-1, 6-3 victory over Saisai Zheng in one hour, 24 minutes.
The sixth-seeded Tomljanovic will take on Tamara Zidansek for a place in the final.
